(a) OsbHLH166 binds directly to the AuxRE of the OsABCB4 promoter, promotes the expression of OsABCB4, increases auxin efflux, and leads to a decrease in auxin content, which results in shorter grain and decrease of grain weight. Meanwhile, OsbHLH166-OsABCB4 negatively regulate GL7 or GS2 expression, positively regulate TGW3 expression, which ultimately leads to shorter grains in their overexpression lines. (b) In osbhlh166 and osabcb4 mutants, the decrease of auxin efflux leads to an increase in auxin content, and altered expression of GL7, GS2 and TGW3 genes, which results in longer grain and increase of grain weight. →, activate; ┴, repress; ↑, increase; ↓, decrease.
