Three-Step Click-Chemistry Method for Rapid PROTAC Assembly (IMAGE)

Researchers used a modular sequence of copper-catalyzed azide–alkyne cycloaddition (CuAAC), sulfur (VI) fluoride exchange (SuFEx), and Michael addition reactions to quickly assemble proteolysis-targeting chimeras (PROTACs) from simple building blocks. This approach simplifies the overall synthesis process and speeds up the development and testing of new protein-degrading drug candidates.
