In part 1, the initial model was selected and given a Trp residue for robust quantitation (along with 5 other mutations), generating the intermediate structure pdb:7KAE. In part 2, structure 7KAE was extended, followed by differentiation to resolve clashes. Here 'clashes' includes unfavorable steric, electrostatic and hydropathy interactions. In part 3, a core Phe residue was added, along with further mutations. The designed structure was then expressed and imaged by diffraction at 1.75 A (pdb:9CFS).
