A Listeria monocytogenes drug delivery system faces numerous challenges including sufficient invasion by payload-bearing LM, the release of cytotoxic payload and delivery of drug at sufficient intracellular concentration for cytotoxicity in target cancer cells. Researchers developed two approaches of Listeria monocytogenes-mediated cytotoxin delivery and release: endolysosome targeting using SN38/Dox-ADCs and cytoplasm targeting using saporin (Figure 1). The efficacy of the LM cytotoxin delivery vehicle approach was tested both in vitro and in vivo in sarcoma and MSS CRC models.
