Graphical illustration depicting the multifaceted role of FAK in cancer. FAK is activated by integrins, RTKs, and GPCRs, driving downstream oncogenic processes including proliferation, EMT/invasion, metastasis, cancer stemness, and chemoresistance. FAK also modulates the tumor microenvironment by regulating endothelial cells, CAFs, macrophages, MDSCs, NK cells, T cells, and dendritic cells, impacting vascularization, TME remodeling, and immune evasion. Emerging FAK-targeted therapeutic strategies include kinase inhibitors, combination therapies, PROTAC degraders, PPI blockers, and natural compounds.
