This study provides the most comprehensive disease-proteome atlas of childhood adversity to date, reveals plasma proteins as key mediators of associations between childhood adversity and disease, and presents a proteomic childhood adversity score that improves disease risk prediction beyond the self-reported measure alone. These findings support a key role for plasma proteins in the biological embedding of childhood adversity and suggest therapeutic potential to reduce long-term disease burden.
