GABPA couples promoter recognition with transcriptional elongation through Integrator recruitment (IMAGE)
Caption
Schematic representation of the dTAG-based acute protein degradation strategy and the temporal consequences of GABPA depletion. In wild-type cells, GABPA binds promoters and associates with the Integrator complex, particularly the catalytic module containing INTS4/9/11, facilitating productive transcription elongation. Upon acute GABPA degradation, promoter accessibility remains largely unchanged, but Integrator recruitment is impaired, leading to reduced Pol II Ser2 phosphorylation and defective transition into productive elongation at ribonucleoprotein biogenesis genes. Prolonged GABPA depletion further compromises global transcriptional programs, resulting in impaired chromatin accessibility and widespread transcriptional defects.
