Schematic overview of the experimental workflow. (A) A total of 90 LUAD patients were recruited in this study. Tumor tissue, ANT, and plasma samples were collected from each patient. All patients were followed for 2 years, and patients with recurrence status, non-degraded samples, and complete follow-up information were included in the final analysis. (B) Total DNA was extracted from the samples, linear DNA was digested using exonuclease, and circular DNA was enriched and purified by rolling circle amplification, followed by 150 bp paired-end sequencing on the NovaSeq 6000 platform. (C) eccDNA was identified using Circle-Map and subjected to downstream analyses.
