RPA was performed in the presence of labeled primers using cytochrome b DNA as a template. RPA generated labeled amplicons that were applied to the lateral flow strips. In the presence of the target DNA, an immune complex of a FAM-labeled terminal with an anti-FAM–GNP conjugate was formed as the labeled amplicons passed along the strip. The following streptavidin–biotin reaction occurred in the test zone and caused its colorization via GNP inclusion.
