Gene Deletions and Duplications Reveal Our Genetic Storyline

膨大な集団全体のコピー数変異を詳細に調べたところ、選択がいかに地球上のヒトのゲノムに影響するかについてより明らかな像が得られた。コピー数変異とは、DNAの多くの部分が複製または削除される時に生じる、ゲノム間の構造上の違いである。これには、多数の遺伝子を含むゲノム領域や、重要な調節領域などが含まれ得る。このため、このような変化は選択下で生じるという仮説が立てられているが、ヒトのゲノム内でこれらの領域を削除したり維持したりする力については十分に理解されていない。これらのパターンをグローバル規模でよりよく理解するために、Peter Sudmantらは、125のヒトの集団から236の個人のゲノムを対象にコピー数変異の分析を行った。この解析の結果、人種によるパターンや、古代のヒト科に由来する混合したゲノムの維持のパターンを明らかにすることができた。例えば、オセアニアの集団では、デニソワ人系列に由来する広範な複製が保持されていた。予想されるように、非アフリカ人集団はより多くの集団ボトルネックを経てきたために多様性のレベルが低いが、これと比べてアフリカ人では祖先のゲノム配列が認められる割合が高かった。こうしたボトルネックイベントの結果として、非アフリカ人ではアフリカ人集団より遺伝子の削除も少なかった。驚くべきことに、この研究結果から、DNAの削除は選択をより反映しているのに対して、DNAの複製は集団ごとの特異的な構造をより強く示していた。こうしたヒトのゲノムにおける違いは、世界レベルでのヒトの移動と定着について、また人類が各世代にわたって直面してきた人口動態的および選択的な圧力についての興味深いストーリーを物語ってくれる。
