研究者らにより、マウスの一回の食餌摂取量の調節において重要な役割を担う脳内の酵素が同定され、この酵素が欠損するとマウスの食餌摂取量が肥満になるまで増加することが明らかにされた。この結果は、ヒトの肥満治療薬の新たな標的を示唆するものである。肥満は数多くの疾患と関連するが、重度の過体重に対して利用できる治療はない。食餌量を調節し、それにより体重を調節する脳内メカニズムについてさらなる知見を得るため、Olof Lagerl�fらはこれまで肥満に関連するとされてきた経路に注目した。特に、O-GlcNAc転移酵素(OGT)は、食物処理に関わるホルモンであるインスリンと相互作用する。研究者らが成獣マウスの脳内で、特に視床下部室傍核(PVN)領域に影響を及ぼすニューロンにおいてOGTをノックアウトしたところ、マウスの体重に劇的な影響が認められた。3週間の間に、マウスの脂肪組織量が3倍に増加した一方で、除脂肪体重は変化しなかった。OGTのノックアウトにより1日当たりの食餌摂取量は急激に増加し、対照マウスの2倍以上の量で平衡状態に達した。OGT欠損マウスの食餌量を対照マウスの摂取量と同量に制限した場合、正常な体重が維持された。さらに調べたところ、食餌摂取活動によって、PVNでOGTを発現するニューロンが活性化されるが、OGTの欠損はこれらのニューロンの活性化を完全に阻害し、ニューロンの発火頻度が大幅に低下することが明らかになった。これに対して、OGTを発現するPVNのニューロンを刺激すると、24時間の累積食餌摂取量が減少した。Gary SchwartzのPerspective記事では、これらの結果についてより詳細に論じている。
