Label-Free 3D Imaging of Lysosomal Remodeling in Niemann-Pick Type C1 Disease Models Using HTFC (IMAGE)

This visual abstract illustrates the power of holographic tomographic flow cytometry (HTFC) in distinguishing healthy (WT) and diseased (NPC1 KO) cells through 3D, label-free imaging of lysosomes (green) and nuclei (blue) in live suspended cells. The technique enables quantitative analysis of lysosomal aggregation and morphology, providing novel biomarkers to evaluate therapeutic efficacy. Treatments with Cyclodextrin or siSPAG9 in NPC1-deficient cells restore lysosomal distribution, moving the cellular phenotype closer to healthy conditions. HTFC offers a promising tool to support diagnosis and personalized treatment strategies for lysosomal storage diseases.
