: FXYD5 and FXYD6 primarily exhibit oncogenic roles by influencing pathways such as TGF-β/SMAD, PI3K/AKT, and NF-κB. Conversely, FXYD2 acts as a tumor suppressor, particularly in ovarian clear cell carcinoma, where it is transcriptionally regulated by HNF1B. FXYD3 demonstrates tumor-promoting and tumor-suppressive roles: it promotes tumorigenesis in cancers like pancreatic cancer and bladder cancer, but exerts suppressive effects in certain conditions. The red denotes tumor-promoting activity, the green indicates tumor-suppressive function, and the yellow highlights proteins with both roles.
