When the transcription factor RpaA (pink and magenta) is modified by addition of a phosphate group, it binds to DNA, which at this promoter blocks the binding of the enzyme (salmon) that transcribes the fluorescent reporter gene. By controlling when RpaA is phosphorylated, the clock turns transcription of this gene off in the day and on at night.
