Immobilized microorganisms are collected on a membrane filter and rinsed with pure water. The hydrated specimen is frozen by gently contacting it with a copper block pre-cooled to -80°C, then mounted on a pre-cooled steel support stage, and transferred to a freeze-dryer. Ice is then removed by sublimation. The workflow can be carried out with either a standard laboratory freeze-dryer or a dedicated freeze-drying apparatus.
