Joint measurement, functional testing, and research applications in brain multi-omics. (IMAGE)

The left column summarizes measurement strategies that capture temporal changes and variation across brain regions and cell types. The middle column presents two approaches to functional testing: multiplexed CRISPR perturbation in human brain organoids, and single-nucleus profiling of resected patient brain tissue combined with in utero electroporation in the embryonic mouse brain. The right column shows applications to disease research and neuroengineering, including early postnatal dorsolateral prefrontal cortex from donors with trisomy 21, postmortem multiregion brain samples used to study Alzheimer's disease, diagnostic and post-treatment tumor samples from patients with pediatric high-grade glioma, and a mouse model implanted with a non-functional intracortical probe. CRISPR, clustered regularly interspaced short palindromic repeats; DLPFC, dorsolateral prefrontal cortex. Credit: Peng Luo and Hongbo Guo.
