Organoid coculture strategies for modeling the TME (IMAGE)
Caption
Organoid coculture systems can be broadly classified into two categories based on how the TME is established. In reconstituted TME models, organoids are first established in Matrigel within growth medium, and then exogenous stromal or immune cells are added to reconstitute specific TME components. These models can be generated using submerged Matrigel coculture, 3D bioprinting, or microfluidic platforms. In native TME models, minced or digested primary tumor tissues are embedded directly in collagen gels and cultured under ALI conditions or within microfluidic devices, thereby preserving the original immune and stromal components alongside the tumor cells without exogenous reconstitution. 3D, three-dimensional; ALI, air-liquid interface; TME, tumor microenvironment.
