Gut microbe-made metabolite UDP-GlcNAc hijacks the host "sugar-switch" (O-GlcNAcylation) to fuel breast tumour progression and chemotherapy resistance (IMAGE)

Graphical Abstract: Ruminococcus lactaris in the gut synthesizes UDP-GlcNAc, which enters the circulation and activates the host enzyme OGT. OGT-mediated O-GlcNAcylation stabilizes transcription factor NFIB, which upregulates OGT and the proto-oncogene KIT, forming a positive-feedback loop that drives breast cancer proliferation, invasion, migration, and paclitaxel resistance. Inhibition of OGT blocks this pro-tumor axis.
