Huashi Baidu formula alleviates lipopolysaccharide-induced inflammation and acute lung injury in mice by targeting nuclear factor κB/phosphatidylinositol 3-kinase and peroxiredoxin 5

This study is led by Professor Jigang Wang and Professor Ping Song (China Academy of Chinese Medical Sciences,Beijing, China). Huashi Baidu formula (HSBD) with the effects of eliminating dampness, clearing heat, ventilating lung, and removing toxin has been proven to be effective in the treatment of COVID-19. The authors combined the cellular thermal shift assay (CETSA) and proteomic analysis to identify the targets of HSBD by taking this formula as a whole. This study provides an approach to characterize the effects of formulas, which can be applied in the research on the TCM treatment mechanisms and the validation of target proteins.

 

The authors established the mouse model of LPS-induced ALI and provided evidence that HSBD could serve as a therapeutic agent to alleviate inflammation and lung injury via NF-κB and phosphatidylinositol 3-kinase (PI3K) signaling as well as peroxiredoxin 5 (PRDX5).  In this study, HSBD was considered as a whole, and the combination of CETSA and proteomic analysis provides an approach to characterize the effects of formula and can be used for the research on TCM treatment mechanisms and the validation of target proteins.

 

In summary, HSBD possesses protective effect against ALI induced by LPS by alleviating oxidative stress and inflammation via NF-κB and PI3K signaling pathways, as well as targeting

PRDX5. The present study provides evidence that HSBD alleviates inflammation and lung injuries and serves as a target protein for the screening of effective ways against infection.
