Evaluation of apoptosis and proliferation of macrophages in vivo. (IMAGE)
Caption
(A) C57BL/6J mice were treated with ConA for 72 hours. Liver sections were stained with IBA1, CLEC4F, KI67 and Cl-CAS3. The left panel showed the representative staining of the four markers. The right panel showed representative images of proliferating MoMFs (1), apoptotic MoMFs (2), proliferating KCs (3) and apoptotic KCs (4) as indicated by arrows. (B) Quantification of the percentage of Ki67+proliferating KCs and MoMFs (n=4 mice) and (C) Cl-CAS3+apoptotic KCs and MoMFs (n=4 mice) in healthy liver and injured liver. Values in B and C are represented as means±SD. ***p<0.001. Cl-Cas3, Cleaved-Caspase3; CLEC4F, C-Type Lectin Domain Family 4 Member F; ConA, concanavalin A; DAPI, 4',6-diamidino-2-phenylindole; IBA1, ionized calcium-binding adaptor molecule 1; KC, Kupffer cell; MoMFs, monocyte-derived macrophages.
