MIB2 constitutively binds and ubiquitylates cFLIP. Upon TNF stimulation, caspase 8 and ubiquitylated cFLIP are recruited to the death-inducing complex. Ubiquitylated cFLIP prevents a proper oligomer formation with caspase 8, thereby suppressing caspase 8 activation and apoptosis.
